DNA methylation patterns in blood split into ten age‑related types A preprint posted 29 August mapped age‑related changes in DNA methylation — chemical tags on DNA — in whole blood. Using 3,501 whole blood samples, the researchers confidently assigned more than 600 000 of the 811 870 CpG sites — short DNA regions where methylation is measured — to ten distinct methylation patterns. For each CpG site they plotted age on one axis and methylation level on the other, letting a neural network recognise the shape of the plot and assign the site to one of the ten types. One of the types was linked to an inflammatory signal and to mortality. The same age‑related methylation shift can look different: some sites show a gradual change in average level, others display increasing variability between people, and yet others form clusters tied to sex or genetic variants. To isolate the component driven by increasing variability, the authors built the NoiseClock metric from 399 CpG selected in three independent cohorts after statistically correcting for the estimated proportions of twelve cell types. At these CpG the average methylation level does not change with age, but the inter‑individual spread grows. NoiseClock was defined as the average deviation of these marks from their usual level and was tested in 14 blood‑based datasets and five sorted immune‑cell datasets, where the age association persisted. NoiseClock also correlated with a methylation‑based inflammation score that had been adjusted for cell composition.
