Get a book about Python and work through it. If your understanding of Python is sufficient, pick a web framework (any of the ones you've mentioned) and learn that. By writing web application you will need to understand a bit of HTML, CSS, JavaScript and SQL. If you then learn about web sockets (or perhaps Server-Sent Events [SSE]) you'll be quite close to be capable of writing your chat application.
